The first white person to live in Hillsborough was John James Audubon who built his cabin on what is now Airport Way in 1847. In 1868, Charles Goodyear established a Mutual Life Insurance Company with office and warehouse space in what is now downtown Hillsborough. Today, Mutual Life Insurance continues as one of the largest insurance companies in Northern California with over $4 billion in assets under management as of September 30, 2018. In 1850, British Gas Light Company founded an oil lamp plant near present-day Hayward Street and Market Street; however this company failed within 3 years due to low production costs and competition from local oil producers. A new gas lighting company was founded by David Sibley called "Kernig Gas" which became successful until 1859 when Kernig merged with another gaslighting company to form "DirecTV". DirecTV was reorganized into two separate companies - American Electric Power (AEP) divisions - each having their own headquarters (Pacific Telephone Building/American Telephone Building) at 2nd Avenue and King Street: ACORN Utilities Incorporated then took over both buildings vacated by DirecTV for other uses after deregulation occurred that year; today Pacific Telephone Building still stands as one of San Francisco's most iconic architecture landmarks (alongside Candlestick Park). Hillsborough also played host to America's first soccer match on October 15th 1886 when 10 teams from New York City participated against 14 teams from California Field Club at Race Track & Sports Grounds off Telegraph Road South; these games were officiated by Judge William McKinney who later became Chief Justice of the Supreme Court of California In 1933, Mayor Ritchie ordered construction began on Interstate 680 which would curves around hills just south of town; this project increased traffic congestion throughout eastern Contra Costa County much like current I-580 freeway will do several decades later while also disrupting community growth patterns along I-680 corridor nearby communities such as Walnut Creek and Hercules

Hillsborough was platted in 1852 by Colonel George Hill and named after his home town in Durham, England. The first post office was opened in 1853, and the first school was built in 1854. The city went through several name changes before settling on its current name in 1867 when it became part of Kern County. In 1905, Hillsborough joined with other cities in what is now Kern County to form the new City of Bakersfield. In 1911, a race riot occurred at Hillsborough; twenty-one people were killed and more than two hundred were injured as hecklers responded to an address by Mayor Edwin Wayman calling for racial integration. The riots are considered to be one of the most deadly incidents during California's "Golden Age" of racing.

The first European to arrive in what is now Hillsborough was a Spanish prospector, José Hidalgo, who discovered gold and silver in 1789. In 1837, a group of Mormon missionaries arrived in what is now Hillsborough and founded the Bishopric of Orange. By 1840, the population of Hillsborough had reached 1000 people. After California became an independent country in 1850, most of the early growth in Hillsborough came from trade with Northern California. The town's first post office was established in 1851. In 1865, when Southern California became part of the United States, Hillsborough incorporated as a city. The city experienced rapid growth during Reconstruction Era due to its strategic location at the mouth of two important rivers: the San Joaquin River and Tampa Bay. However, during World War II, Hillsborough struggled with wartime restrictions on manufacturing that forced it to import goods from outside its borders. During the war years, some residents began to form their own groups to resist military draftsmanship and integration into American society; these groups were known as "Hillsbourgh Defense League". As a result of this resistance, many businesses closed down or relocated within the city limits during postwar years.
