It is the most populous city in Contra Costa County and the second-most populous city in Northern California after San Francisco. The name "Pleasanton" comes from a post office called Pleasant Plains which opened on October 17, 1853. The city was incorporated on November 3, 1875. It is named for Capt. John Condon of the 24th Regiment of volunteers who served during the Mexican-American War and are buried at Mount Diablo Cemetery thereabouts. The old town of Pleasanton was located east of what is now downtown Pleasanton and included portions of what are now Norwood and Irvington areas. The new town was founded by John Sutter on May 12, 1879 as a result of his donation of land to the Town Trustees for $5 per acre (equivalent to $ ). In 1849, a group of farmers in what is now Pleasanton started to build a cabin on the bluff overlooking the San Joaquin River. The name of the settlement was first given to it by an early settler, Samuel Morse, who named it for his home town of Pittsfield, Massachusetts. The first post office was established in 1852 and soon became a center for wagon trains moving westward from Sacramento and other points along the California Trail. By 1865, Pleasanton had become an important railroad junction and trade center. Growth continued until the Great Depression hit in 1930 when many businesses closed their doors because of difficult economic times. In 1941, during World War II, Pleasanton played a key role as one of three U.S. military bases located in Contra Costa County. After the war ended, many businesses reopened but growth slowed again due to further recessionary conditions that began in the mid-1970s. There has been recent progress however with several new restaurants and stores opening up around town since 2000 including local brewery Stone Brewing Company which built its current brewery building in 2006 adjacent to Buster's Donuts & Coffee shop on Main Street.

In 1873, a railroad station was built in the town, and in 1876, a water tower was erected. The first school opened in Pleasanton on July 4, 1876. In 1915, a small dam was built north of the town to flood land that had been taken from the Sacramento River for irrigation purposes. In 1923, Pleasanton became an incorporated city with a population of about fifty people. It experienced rapid growth during World War II due to its status as an important manufacturing center for munitions production. After the war ended, however, crime rates began to increase and many businesses closed down; consequently, downtown business began to decline until it regressed after 1990 when new development began increasing activity on campus and along Main Street
